Service Description

DeWalch offers a full length insert sequencing service for large insert vectors such as BACs, cosmids and fosmids. BAC end sequencing with gene-specific or vector-specific primers is available including full gene DNA sequence analysis. Our service includes colony picking, bacterial cell growth, DNA extraction from clones, DNA sequencing for a single sample to a 96-well platform for high-throughput scale processing.

Sample Requirements:

  • DNA sample in Tris buffer or molecular grade water only!
  • DNA sample in tubes or 96 well plate
  • 2 µg of DNA at 250 ng/µl required
  • Customer primer (if not the universal primer) needed

BAC End-Sequencing ABI 3130xl Genetic Analyzer automated fluorescent DNA cycle sequencing of a Xenopus BAC clone purified using DeWalch's NeXprep Ultra Pure Plasmid Kit. Using ABI KB basecaller software (ABI DNA sequencing analysis software V5.2), the QV20 score of this particular sequence is 961 bases.

BAC End-Sequencing Results Sequencing results of BAC clones using DNA purified by the DeWalch NeXprep Ultra Pure Plasmid Purification Kit. DNA sequencing was performed using AB (Applied Biosystems) Big Dye Terminator v3.1 Cycle Sequencing Kit each with a 1/8 X reaction. Data was analyzed using AB DNA Sequencing Analysis Software V5.2.
